Over the last few months we’ve had questions from the community on what the best input DNA concentration is when expressing protein, such as GFP, in liposomes. As we’ve been validating and tuning modules for the Detector Cells this question has come up for us as well.
Takeaways
- A wide range of DNA concentrations are effective, from <1 ng/uL to 100 ng/uL.
- GFP expression is broadly correlated to input DNA concentration, although the relationship is not perfectly linear.
- At high concentrations, we see a bimodal effect appear. At many concentrations, liposomes appear to burst or leak, which may be a result of membrane permeation, or may be a direct effect of protein expression.
- The liposome analysis toolkit is an effective way to measure expression in liposomes at scale.
